Orforglipron is a once-daily oral GLP-1 receptor agonist from Eli Lilly. It is a small molecule, not a peptide, and is sold as Foundayo.
Orforglipron activates the same GLP-1 receptor as semaglutide and tirzepatide, but it is a synthetic chemical compound rather than a chain of amino acids. Lilly states that Chugai Pharmaceutical discovered it and that Lilly licensed it in 2018. It is taken as a tablet, and the FDA label allows dosing with or without food. The label also notes that orforglipron is not pharmacologically active in rats or mice and did not produce tumors in rodents; Foundayo still carries the class boxed warning for thyroid C-cell tumors.
The main obesity trial was ATTAIN-1 (NEJM, 2025). It randomized 3,127 adults with obesity and without diabetes to orforglipron 6 mg, 12 mg or 36 mg daily, or placebo, for 72 weeks. Under the treatment-regimen estimand, mean body weight fell 7.5%, 8.4% and 11.2% in the three dose groups, versus 2.1% on placebo. Adverse events led 5.3% to 10.3% of orforglipron patients to stop treatment, compared with 2.7% on placebo, mostly because of gastrointestinal effects.
Yes. The FDA approved orforglipron as Foundayo on April 1, 2026, for chronic weight management in adults. It is not approved for type 2 diabetes.
The Drugs@FDA record lists Foundayo as a new molecular entity under NDA 220934. The approved use is adults with obesity, or adults with overweight plus at least one weight-related condition, alongside a reduced-calorie diet and more physical activity (Lilly Medical). As of October 2026, Lilly describes orforglipron as still being studied for type 2 diabetes, and Drugs@FDA shows no diabetes approval.
The genuine product is a pharmacy tablet. Any powder or capsule sold as research orforglipron is not Foundayo and has no FDA oversight. Because a white powder looks the same whatever it contains, a credible result for such a batch identifies the compound by mass spectrometry and reports how much is present per unit, rather than giving a purity figure alone.
Before trusting a label, check that batch’s published result on our latest tests page or in Janoshik’s public results database. No results are published for orforglipron in our index yet. When the first published result arrives, a vendor leaderboard appears above.
